Not sure if directly relevant to your current work, but I remember being very impressed when this paper was presented a couple of years back at ACA - demonstrating how careful optimization could drastically improve SEM contrast on multiple instruments (IIRC) www.sciencedirect.com/science/arti...

De novo tilt series alignment accuracy is still the #1 problem, even with thinner samples. Can't recognize particles if they're mangled by misalignment.
