While screening for motifs, we discovered that locally scrambled IDRs were functional, and found conformational dynamics rather than linear sequence predicted function. We then used IDR dynamics as a design target and generated functional IDRs de novo.

Here, we develop a system to map sequence to function in the IDR of RUNX2, the transcription factor that regulates bone development. We show that specific IDR function is encoded beyond exact amino acid sequence order.

Folded protein design is now routine, but ~35% of the human proteome lacks stable 3D structure. Synthetic intrinsically disordered regions (IDRs) could be used for cell engineering. However, without structure, the target for biologically functional disordered protein design is not obvious.
