@agatakstepien.bsky.social2. Interestingly, another clue lies in the cleavage-termination distance, which is particularly short in primary tumors, especially at oncogenes. The shorter the distance, the higher the nuclear gene expression. This relationship also holds in cervical and pancreatic cancer cells.
Show Post1. And the differences don’t end there! We also observed contrasting patterns of alternative polyadenylation and transcription termination during colorectal cancer progression. Remarkably, metastatic cells partially revert toward the termination patterns seen in healthy cells.
Show Post3. Cleavage-termination distance is also linked to sensitivity to changes in #PCF11 and #CPSF73 levels. Could this differential dependence on pre-mRNA 3'end processing and termination factors reveal an overlooked vulnerability in tumor cells and open new avenues for cancer therapy?
Show Post4. Finally, congratulations and a big thank you to @stoplab.bsky.social, @agatakstepien.bsky.social, Martyna Plens-Gałąska, Magda Kopczyńska, Kaludia Ostatek, Mariia Kapeliukha, Shazia Irshad, Hiroshi Kimura, Michal and Stephan Hamperl for their great collaboration on this project.
Show PostPrimary and metastatic colorectal tumor cells share a common origin, but do they share the same molecular vulnerabilities? Surprisingly, primary cells depend far more on #PCF11 and #CPSF73, two key pre-mRNA 3' end processing/termination factors. Our latest study explores why: tinyurl.com/3pkeuctp
