Thrilled to share my PhD paper showing how SUMO regulates PRC1 clustering and 3D genome organisation independently of H3K27me3! Grateful to @djost-physbiol.bsky.social group and the Fabian Erdel group for this great collaboration. doi.org/10.1093/nar/...

It provides a simple framework to understand the regulation of Rad51 filament structure and function by RAD51 paralogs complexes and functional homologs of Srs2.

Show Post

Through computational modeling, we show that such outcome can be achieved if Rad55-Rad57 restricts Srs2 disruption activity to individual Rad51 segments. It results in the “pruning” that allows surviving ones to grow longer. Reduced filament segmentation = increased stiffness!

Show Post

Through in vitro reconstitution experiments, we surprisingly find that the disruption activity of Srs2 drives the assembly of long and stiff Rad51 filaments when Rad55-Rad57 is present. How can that be?

Show Post

Importantly, by mapping genome-wide the contacts made by Rad51 filaments and by quantifying D-loop products, we show that the combined presence of Rad55-Rad57 and Srs2 is required for homology search to turn genome-wide; a switch that requires assembly of long and stiff Rad51 filaments.
