Unfortunately, this is something that will require different art for each color. If you look at other puzzle games, that's what they do. For example, here's Puzzle Quest, which puts a different little glyph on each color:

Looks interesting! I can't quite say what kind of game it is, maybe PvP tetris? For a second I thought the two characters might be fighting each other, but since there's no health bar I'm not so sure. I also have no guess as to why the tetris part is stacked the way it is :)

Oh nice, a Puzzle Fighter game! Good luck with development! Readibility-wise, vary the different pieces by both color AND shape, or else the game isn't accessible to colorblind people. You can find colorblind filters that show you what you game looks like to different folks.
