When imaging at high mag using cryo-ET, field of view reduces. How to overcome this limit? How do we get a "bird’s-eye view" without compromising resolution? Our very own @HamishBrown has a solution to this: “montage tomography with a rectangular beam!” www.biorxiv.org/content/10.6...

I'd vote for the future where there's the option of a box that pops up when lazy nonsense appears and says "this looks like AI slop. Would you like me to find a piece written by a thoughtful human on this topic - something that's valued by other humans and is actually worth reading?"

Me to blame for the up/down view, but we gonna stay with it! 🤪 I do feel dizzy when inverting the thing... Probably, comes from previous SecYEG projects - there, cytoplasm/targeting is commonly drawn above the membrane, so things go from top to the bottom

In early days we tried to isolate the ring (in absence of PelB), but pulled only small fragments. Also it crystallizes commonly as dimers/trimers, so the barrel (and the periplasmic domain of that) is needed to hold it together.

And for the assembly pathway, my guess is that PelB is inserted on its own via BAM. PelC's (delivered by Lol route) initially diffuse at the membrane either as monomers or small oligomers, and dock on the barrel later.
